Therefore, in this study, we used human liver cancer cell SK-Hep-1 cells with high metastatic activity as an experimental model to explore the migration, invasion, adhesion, and MMPs activity and performance of Hispolon, a component of Phellinus linteus, on SK-Hep-1 cells. the influence. The Transwell chamber method was used to evaluate the effect of Hispolon on the adhesion, migration and invasion abilities of SK-Hep-1 cells. Gelatin zymography was used to analyze extracellular MMP-9 and MMP-2 activities. The protein expression of MMP-9 and MMP-2 in cells was analyzed by Western blotting method. The results showed that after co-culture with Hispolon (0, 1, 2.5, 5, 7.5, 10, 25 and 50 μg/ml) for 24 and 48 hours, SK-Hep-1 cells at 24 hours at concentrations of 1, 2.5 and 5 , 7.5 and 10 μg/ml are not cytotoxic, so non-cytotoxic concentrations and time points were selected for subsequent experimental studies.
